醫(yī)學(xué)論文范文:雌激素及拮抗劑三苯氧胺對子宮內(nèi)膜癌細(xì)胞生長的影響
【摘要】 目的 研究雌激素及拮抗劑三苯氧胺對子宮內(nèi)膜癌細(xì)胞HEC1B生長的影響。方法 體外培養(yǎng)中分化子宮內(nèi)膜癌細(xì)胞HEC1B,按實(shí)驗(yàn)?zāi)康姆謩e加入17β雌二醇(E2,10-8M)、三苯氧胺(TAM,10-7 M)刺激24、48、72、96 h后,采用四甲基偶氮唑藍(lán)(MTT)比色方法比較各個條件下的細(xì)胞數(shù)目。用流式細(xì)胞術(shù)方法檢測加入E2、TAM后的細(xì)胞周期變化,比較各個條件下的S期細(xì)胞比率(SPF)。結(jié)果 MTT結(jié)果:E2、TAM在作用72 h和96 h后,可顯著刺激HEC1B細(xì)胞生長(P<0.05)。HEC1B細(xì)胞加入E2刺激后,SPF在加藥72 h和96 h均增高。TAM作用細(xì)胞后,SPF值在藥物作用48 h起均高于對照組。結(jié)論 雌激素、三苯氧胺均對子宮內(nèi)膜癌HEC1B細(xì)胞有促生長作用。

【Abstract 】 Objective To investigate the effect of estrogen and tamoxifen on the growth of endometrial carcinoma cell line-HEC1B. Methods The endometrial cancer cell lineHEC1B was cultured in vitro. The cells were stimulated with 17βestrodiol(10-8M)and tamoxifen (10-7M)for 24 h,48 h,72 h,96 h respectively. The proliferation capacity of the cells was evaluated by means of mircocutrue tetrazolium assay (MTT) and the changes of cell cycle after E2 and TAM introduction were detected by flow cytometry (FCM).The cells percentage in S phase(SPF)in different conditions was compared.Results Both 17βestrodiol and tamoxifen could significantly stimulate the growth of HEC1B cell 72h and 96h after E2,TAM introduction. HEC1B cells could increase SPF 72h and 96h after E2 stimulation. The SPF of HEC1B cell 48h,72h,96h after TAM introduction was higher than that of control group.Conclusion Both estrogen and tamoxifen can stimulate the growth of HEC1B cell line.

子宮內(nèi)膜癌是女性生殖道最常見的三大惡性腫瘤之一,近年來其發(fā)病率呈上升趨勢,占女性生殖系統(tǒng)惡性腫瘤的20%~30%[1]。目前普遍認(rèn)為子宮內(nèi)膜癌是激素依賴性腫瘤之一。其發(fā)生、發(fā)展與多種激素及其拮抗劑、受體密切相關(guān)。本實(shí)驗(yàn)研究雌激素及其拮抗劑三苯氧胺(TAM)對子宮內(nèi)膜癌細(xì)胞生長的影響醫(yī).學(xué).全.在.線bhskgw.cn。
1 材料與方法
1.1 實(shí)驗(yàn)材料 選用中分化子宮內(nèi)膜癌細(xì)胞系HEC1B,由北京大學(xué)第一醫(yī)院婦產(chǎn)科研究室傳代。
