本發(fā)明涉及一種P53融合蛋白。本發(fā)明將人wtP53基因cDNA片段和蛋白傳導(dǎo)結(jié)構(gòu)域(PTD)融合,插入到大腸桿菌原核表達(dá)載體PGEX-3X中,得到重組表達(dá)質(zhì)粒PGEX-3X-P53,利用IPTG誘導(dǎo)產(chǎn)生wtP53融合蛋白;分子量約為53kbp,經(jīng)DNA序列測(cè)定,它由1218個(gè)核苷酸組成;經(jīng)MTT法及TUMEL法體外實(shí)驗(yàn),表明wtP53融合蛋白可明顯抑制肺癌細(xì)胞H1299和肝癌細(xì)胞HePG2的增殖,促進(jìn)其調(diào)亡,本發(fā)明為應(yīng)用wtP53融合蛋白治療腫瘤提供了可靠的途徑。
